RESUMO

Islet Neogenesis Associated Protein pentadecapeptide (INGAP-PP) increases ß-cell mass and function in experimental animals. A short clinical trial also yielded promising results. However, HTD4010, a new peptide derived from INGAP-PP, was developed in order to optimize its specific effects by minimizing its side effects. To study and compare the tertiary structure, stability dynamics, and plasma stability of HTD4010, an INGAP-PP analogue. Both peptides were pre-incubated in human, rat and mouse plasma at 37 °C, and their presence was identified and quantified by high performance liquid chromatography at different time-points. GROMACS 2019 package and the Gromos 54A7 force field were used to evaluate overall correlated motion of the peptide molecule during molecular dynamics simulation by essential dynamics. HTD4010 exhibited significantly larger plasma stability than INGAP-PP, and its structural stability was almost 3.36-fold higher than INGAP-PP. These results suggest that HTD4010 may facilitate longer tissue interaction, thereby developing higher potential biological effects. If so, HTD4010 may become a promising therapeutic agent to treat people with diabetes. Communicated by Ramaswamy H. Sarma.

RESUMO

Phased national medical licensing examination requires clinical students to have the ability to apply basic medical knowledge to solve comprehensive clinical problems. But the traditional teaching patterns of Medical Microbiology experiments can no longer meet requirements of actual education effect. In order to accommodate new situation, we have integrated medical licensing examination with microbiology experimental teaching syllabus and contents. We have also comprehensively applied a variety of teaching methods and curriculum formative assessment in class. These reforming methods have efficiently improved classroom teaching effect and greatly inspired the enthusiasm of students, and effectively improved early clinical thinking ability and innovation ability of students.

BACKGROUND: This study aims to explore the changes in functional neuroimaging in bipolar depression patients with anxiety symptoms (BDP-A). METHODS: Forty-five BDP-A patients, 22 bipolar depression patients without anxiety symptoms (BDP-NA), and 48 healthy controls (HC) were finally involved. The low-frequency oscillation characteristics, functional connectivity (FC), and network properties among the three groups of participants were analyzed. RESULTS: Compared with the BDP-NA group, BDP-A patients exhibited significantly decreased amplitude of low-frequency fluctuation (ALFF) in the left middle frontal gyrus (MFG), superior occipital gyrus, and inferior parietal, but supramarginal and angular gyri (IPL). Enhanced FC from left IPL to middle temporal gyrus, from left precentral gyrus (PreCG) to bilateral angular gyri, medial superior frontal gyrus, and left superior frontal gyrus (SFG)/MFG were also revealed. Compared with HC, the BDP-A group showed remarkably increased ALFF in the left MFG/PreCG, right superior parietal gyrus, while decreased ALFF in the left inferior frontal gyrus, opercular part, and SFG. In addition, higher regional homogeneity in the left MFG/PreCG was found. LIMITATIONS: The limitations are as follows: (1) relatively small sample size; (2) not all the patients were drug-naive; (3) lack of pure anxiety disorder patients as a controlled group; (4) mental health conditions of HC were not systemic evaluated. CONCLUSIONS: BDP-A patients showed significant differences in resting-state fMRI properties when compared with BDP-NA or HC group. These results may infer the dysfunction of the dorsal attention network, the default network, and the fronto-limbic system as well as disrupted brain network efficiency in BDP-A patients.

RESUMO

COVID-19 is caused by the SARS-CoV-2 coronavirus and was first reported in central China in December 2019. Extensive molecular surveillance in Guangdong, Chinas most populous province, during early 2020 resulted in 1,388 reported RNA positive cases from 1.6 million tests. In order to understand the molecular epidemiology and genetic diversity of SARS-CoV-2 in China we generated 53 genomes from infected individuals in Guangdong using a combination of metagenomic sequencing and tiling amplicon approaches. Combined epidemiological and phylogenetic analyses indicate multiple independent introductions to Guangdong, although phylogenetic clustering is uncertain due to low virus genetic variation early in the pandemic. Our results illustrate how the timing, size and duration of putative local transmission chains were constrained by national travel restrictions and by the provinces large-scale intensive surveillance and intervention measures. Despite these successes, COVID-19 surveillance in Guangdong is still required as the number of cases imported from other countries is increasing. HighlightsO_LI1.6 million molecular diagnostic tests identified 1,388 SARS-CoV-2 infections in Guangdong Province, China, by 19th March 2020 C_LIO_LIVirus genomes can be recovered using a variety of sequencing approaches from a range of patient samples. OBJECTIVE:To investigate the protective effect an d possible m echanism of Shenqi zhilong decoction (SZD) combined with Tripterygium polyglycoside tablets (TPT)on membranous nephropathy (MN)model rats. METHODS :MN rat model was established by subcutaneous and caudal vein injecting cationic bovine serum albumin + incomplete Freund adjuvant emulsion for 6 weeks. At the 3rd week of modeling ,model rats were randomly divided into model control group ,SZD low-dose and high-dose groups (4,8 g/kg,by total crude drugs ),TPT group (9 mg/kg),low-dose and high-dose of SZD+TPT groups (same dose as single group ),with 10 rats in each group according to 24 h UTP and weight. Another 10 rats withoutmodeling were taken as blank control group. Blank control group was given equal amount of water intragastrically administration groups were given relevant medicine intragastrically,once a day ,for consecutive 4 weeks. The 24 h UTP of rats were detected one day before the last administration;1 h after the la st administration ,blood routine 中indexes(WBC,RBC,PLT),liver func tion indexes (TP,ALB,AST,ALT),blood lipid indexes (TC,TG,HDL-C,LDL-C), the contents of glucose (GLU),urea nitrogen (BUN)and serum creatinine (Scr)were detected in each group. Uranyl acetate-lead citrate staining was used to observe ultrastructural changes of renal tissue. Immunohistochemical method was used to detect the protein expression of TGF-β1 and HPA- 1 in renal tissue. RESULTS :Compared with blank control group ,in the model control group,the glomerular podocytes were widely fused or disappeared ,microvilli were formed ,basement membrane was heavily thickened,a large number of electron dense substance was deposited under the epithelium ,TGF-β1 and HPA- 1 positive cells were significantly increased ;24 h UTP ,PLT,the contents of TC ,TG and HDL-C ,the percentage of TGF-β1 and HPA- 1 positive cells were increased significantly ,while RBC ,the contents of TP and ALB were decreased significantly (P<0.05 or P<0.01). Compared with model control group ,above ultrastructural changes of administration groups were improved to different extents ,and TGF-β1 and HPA- 1 positive cells were decreased. The 24 h UTP ,the percentage of TGF-β1 positive cells (except for SZD low-dose group),WBC(except for SZD alone groups and combination groups ),PLT and TC content (except for TPT group ),TG content (except for SZD low-dose alone and its combination group ),the percentage of HPA- 1 positive cells were decreased significantly ; the percentage of TGF-β1 positive cells in SZD high-dose+TPT group as well as the percentage of HPA- 1 positive cells in SZD+TPT groups were significantly lower than TPT group. RBC and GLU content (except for SZD low-dose group and TPT group ),TP and ALB content (except for SZD low-dose group )were increased significantly ,while the content of GLU in SZD high-dose+TPT group was significantly higher than TPT tablets group (P<0.05 or P<0.01). CONCLUSIONS :SZD combined with TPT can relieve myelosuppression caused by TPT ,reduce proteinuria of MN model rats and improve pathological damage of renal tissue in rats. Its mechanism is related to the down-regulation of protein expression of TGF-β1 and HPA- 1,and the reduction of TC and TG content in the blood.

Solid polymer electrolyte (SPE) is an important part of printed electrochemical gas sensors and are of value to electrochemical sensors. Here, a new type of SPE was prepared by dissolving a poly-vinylidene fluoride (PVDF) matrix in a 1-methyl-2-pyrrolidone (NMP) to immobilize 1-ethyl-3-methylimidazolium tetrafluoroborate ([EMIM] [BF4]), which was then used in a new electrochemical amperometric nitrogen dioxide sensor. The SPE was coated on a single electrode and attached to the electrode to construct a simple two-layer structure. Nitrogen dioxide in the air was reduced on the working electrode at a bias voltage of -500 V. We controlled the components and process parameters separately for control experiments. The results show that the SPE based on [EMIM] [BF4], NMP, and PVDF coated on the electrode at a thickness of 1.25 mm with a 1:1:4 weight ratio under heat treatment conditions of 80°C for 2 min has the best sensitivity. The FTIR and XPS results indicated that SPE is prepared via physical miscibility. The SEM and XRD results showed that the sensitivity of the sensor is strongly dependent on the interconnected pore structure in SPE, and the pore structure is related to the synthesis ratio, morphology, and heat treatment mode of SPE. Moreover, the sensor sensitivity has a certain relationship with SPE conductivity. The reaction principle and cycle performance of the sensor were also studied.

OBJECTIVE: To study the hypoglycemic effect and mechanism of Zicui yin on type Ⅱ diabetic rats, and to provide theoretic basis for clinical application. METHODS: Eighty male rats were fed with high fat/sugar diet for 4 weeks, and then given intraperitoneal injected with streptozotocin (35 mg/kg) to induce type 2 diabetic model. Other 10 rats were included in normal group. Model rats were randomly divided into metformin group (positive control, 0.2 g/kg), model group (constant volume of distilled water), Zicui yin high-dose, medium-dose, low-dose groups (14.0, 7.0, 3.5 g/kg), with 10 rats in each group. After 2 weeks of continuous intragastic administration, the fasting blood glucose of rats in each group was measured by automatic blood glucose meter. The serum insulin level was determined by ELISA. mRNA expression of JNK, Akt and IRS-1 were detected by RT-PCR; The phosphorylation of JNK, Akt and IRS-1 proteins in the pancreatic tissue of rats in each group was detected by Western blot method. RESULTS: Compared with normal group, the fasting blood glucose, mRNA expression of JNK, The phosphorylation of JNK and IRS-1 proteins in the pancreatic tissue of the model group were significantly increased (P<0.05 or P<0.01), while the serum insulin content, mRNA expression of Akt and IRS-1, the phosphorylation of Akt protein were significantly decreased (P<0.01). Compared with model group, the fasting blood glucose of rats in metformin group and Zicui yin high-dose and middle-dose groups decreased significantly (P<0.05), the serum insulin content of rats in both metformin group and Zicui yin high-dose group increased significantly (P<0.01), and mRNA expression of JNK in pancreatic tissue of rats in metformin group and Zicui yin groups decreased significantly (P<0.01), while mRNA expression of Akt and IRS-1 increased significantly (P<0.01). The phosphorylation of JNK and IRS-1 proteins in metformin group and Zicui yin high-dose group decreased significantly (P<0.01), while the phosphorylation of        Akt protein were increased significantly (P<0.01). CONCLUSIONS: Zicui yin can significantly reduce blood glucose level, the mechanism of which may be related to decreasing the phosphorylation of JNK and IRS-1 proteins and increasing the phosphorylation of Akt proteins in pancreatic islets.

Objective:To observe immunological indexes,the quantity of cytokine expression and clinical curative effect of umbilical cord mesenchymal stem cells between before and after the treatment of systemic lupus erythematosus patients.Methods:Selected 10 cases of SLE,on the basis of glucocorticoid and immune inhibitor treatment,intravenous injection UC-MSC of cultivating proliferation within 6 generations.Before and after treatment of UC-MSC testing the relative quantity of cytokine of CTLA-4,IL-15,IL-2,CD86,IL-17c,Foxp3,TGF-β2 which were related of immunopathogenesis of SLE.Before and after treatment to determined SLE disease activity index (SLEDAI) score and detection of blood in the urine routine,liver and kidney function,24 hours urinary protein quantitative,immunoglobulin and complement levels.Results:After treatment the relative expression value of IL-15 and IL-2 was decreased,CTLA-4 was risen.There had no significant difference with the relative expression value of CD86,IL-17c,Foxp3,TGF-β2 in before and after treatment of UC-MSC.After treatment serum complement C3 and C4 level,serum albumin,were risen.24 hour proteinuria and SLEDAI were decreased.There was no serious adverse reaction occurred,no complications related to transplantation in 10 cases.Conclusion:UC-MSC can regulate the expression of cytokines of participate in the immune response in the patients with SLE.Treatment of SLE by UC-MSC can elevate serum albumin and C3 and C4 level,reduce the 24 hours urinary protein quantity,relife kidney damage,improve clinical symptoms;UC-MSC transplantation in patients with SLE have good security;UC-MSC transplantation may be a feasible method for the treatment of SLE.

Objective To investigate the expression of microRNA (miRNA)-3620 in the plasma of patients with anti-tuberculosis drug-induced hepatotoxicity (ATDH).Methods A total of 35 patients with ATDH and 35 non-ATDH paired individuals were included in this study.Plasma miRNA-3620 levels were detected using real-time Polymerase chain reaction.Comparison between two groups was done with t test.Receiver operation characteristic (ROC) curve analysis was performed to determine the diagnostic value of miRNA-3620 in ATDH.Results The relative expression of plasma miRNA-3620 of patients with ATDH and non-ATDH were 1.65±1.43 and 0.71±0.45, respectively, with significantly statistical difference (t=3.703, P<0.01).The cut off value of miRNA-3620 expression was 1.15 and the area under ROC curve were 0.71(95% CI: 0.43-1.45).Based on this cutoff value, the sensitivity and specificity of miRNA-3620 in diagnosing ATDH were 60.0% and 82.9%, respectively;the positive predictive value was 77.8% and the negative predictive value was 67.4%.Twenty-one ATDH cases and 29 non-ATDH cases was correctly diagnosed, with the accuracy of 71.4%.Conclusion The expression of miRNA-3620 in plasma is significantly increased in ATDH patients.

Objective To analyse the characteristics of road traffic accident deaths in Haikou city, and to provide reference for the identification of causes of death and the preventive measures. Methods Totally 498 road traffic accident deaths accepted by the Traffic Police Branch of Haikou City Public Security Bureau in 2014—2016 were collected, and the related parameters such as sex, age, time of the acci-dents, travel mode of the victims, the types of vehicle and the cause of death were analysed. Results Most victims aged 21-40 years old with the sex ratio of 3:1, and the accidents mainly happened in March, April, May and October and peaked at 6:01-8:00 and 20:01-22:00 per day. Riding motorbike and electric bicycle, as travel modes, had the highest accident incidence (30.9%). The vast majority of involved vehicles were motorbike and electric bicycle (57.4%). The most common cause of death was craniocerebral injury, followed by chest and abdominal injury. Conclusion The autopsy of road traffic accident deaths plays an important role in identification of death manner and responsibility confirmation.

Objective To observe the effect of shRNA interference lentivirus vector targeting rat Sirt1 gene on the expression of Sirt1 in retinal ganglion cell (RGC). Methods Four short hairpin (sh) RNA interference sequences targeting rat Sirt1 gene were designed. The target sequences of Oligo DNA were synthesized and annealed to double strand DNA, which was subsequently connected with pGLV3 lentivirus vector to build the lentiviral vector. The positive clones were identified by polymerase chain reaction (PCR) and DNA sequencing. The lentiviral vector construct and lentiviral packaging plasmids were co-transfected into 293T cells, then the titer of lentivirus were determined. The RGC were divided into 6 groups including blank group, negative control group and si-Sirt1-1, si-Sirt1-2, si-Sirt1-3, si-Sirt1-4 groups. Real-time PCR and Western blotting were used to detect the expression of Sirt1 mRNA and protein in the RGC cells. Results PCR and DNA sequencing analysis confirmed that the shRNA sequence was successfully inserted into the lentivirus vector. The concentrated titer of virus suspension was 8×108 TU/ml after the recombinant lentiviral vector successfully transfected and harvested in 293T cells. Comparing with NC group, the expression of Sirt1 mRNA and protein were significantly decreased in the si-Sirt1-1, si-Sirt1-2, si-Sirt1-3 and si-Sirt1-4 groups (F=27.682, 1185.206; P=0.000, 0.000). The si-Sirt1-2 group had the strongest effect in reducing the expression of Sirt1 mRNA and protein. Conclusion The 4 lentiviral vectors harboring RNAi targeting rat Sirt1 gene can effectively down regulate the expression of Sirt1 mRNA and protein in RGC cells.

Objective To analyze the factors influencing the occurrence of macrosomia so as to provide a theoretical basis for the prevention of macrosomia.Methods The data were selected from the findings of Survey on Prevalence and Risk Factors for Birth Defects in Shaanxi Province.This study intended to select data of macrosomia in Xianyang City and its influencing factors for statistical analysis.The rates were compared by χ2 ;the influencing factors were analyzed by univariate and multivariate Logistic regression.Results The incidence of fetal macrosomia among single live birth neonates in Xianyang was 5.2%.The incidence of macrosomia was 6.5%in males and 4.2% in females,with a significant difference (P <0.001).From 2010 to 2013 the annual incidence rate of macrosomia was 5.4%,6.2%,5.9% and 4.4%,with no significant difference (P =0.151).The incidence rate of macrosomia in urban and rural areas was 6.3% and 5.5%,respectively,without significant difference (P =0.556).The incidence of macrosomia was 3.2% in Binxian County,5.6% in Changwu County,6.7% in Liquan County,5.4% in Qianxian County,8.0% in Wugong County,and 4.2% in Xunyi County,with significant differences (P <0.001).Logistic regression analysis showed that the occurrence of macrosomia was related to the baby's sex,mother's age,the times of pregancy,the time of antenatal care,alcohol intake during pregnancy and father's occupation.Conclusion The incidence of macrosomia of Xianyang from 2010 to 2013 was close to the national average.The sex of baby,mother's age,the times of pregnancy,the times of antenatal care,drinking history during pregnancy and father's occupation may affect the occurrence of macrosomia.

Traditionally, the factorization method is applied to reconstruct the 3D geometry of a target from its sequential inverse synthetic aperture radar images. However, this method requires performing cross-range scaling to all the sub-images and thus has a large computational burden. To tackle this problem, this paper proposes a novel method for joint cross-range scaling and 3D geometry reconstruction of steadily moving targets. In this method, we model the equivalent rotational angular velocity (RAV) by a linear polynomial with time, and set its coefficients randomly to perform sub-image cross-range scaling. Then, we generate the initial trajectory matrix of the scattering centers, and solve the 3D geometry and projection vectors by the factorization method with relaxed constraints. After that, the coefficients of the polynomial are estimated from the projection vectors to obtain the RAV. Finally, the trajectory matrix is re-scaled using the estimated rotational angle, and accurate 3D geometry is reconstructed. The two major steps, i.e., the cross-range scaling and the factorization, are performed repeatedly to achieve precise 3D geometry reconstruction. Simulation results have proved the effectiveness and robustness of the proposed method.

<p><b>BACKGROUND</b>The risk of radial artery occlusion (RAO) needs particular attention in transradial intervention (TRI). Therefore, reducing vascular occlusion has an important clinical significance. The aim of this study was to determine the appropriate puncture site during TRI through comparing the occurrence of RAO between the different puncture sites to reduce the occurrence of RAO after TRI.</p><p><b>METHODS</b>We prospectively assessed the occurrence of RAO in 606 consecutive patients undergoing TRI. Artery occlusion was evaluated with Doppler ultrasound in 2 days and 1 year after the intervention. Risk factors for RAO were evaluated using a multivariate model analysis.</p><p><b>RESULTS</b>Of the 606 patients, the RAO occurred in 56 patients. Compared with TRI at 2-5 cm away from the radius styloid process, the odds ratio (OR) for occlusion risk at 0 cm and 1 cm were 9.65 (P = 0.033) and 8.90 (P = 0.040), respectively. The RAO occurred in the ratio of the arterial diameter to the sheath diameter ≤1 (OR = 2.45, P = 0.004).</p><p><b>CONCLUSION</b>Distal puncture sites (0-1 cm away from the radius styloid process) can lead to a higher rate of RAO.</p><p><b>TRIAL REGISTRATION</b>ClinicalTrials.gov, NCT01979627; https://clinicaltrials.gov/ct2/show/NCT01979627?term = NCT01979627 and rank = 1.</p>

RESUMO

Prostaglandin (PG) E2 is an active substance in pathological and physiological mechanisms, such as inflammation and pain. The in vitro high-throughput assay for screening the inhibitors of reducing PEG2 production is a useful method for finding out antiphlogistic and analgesic candidates. The assay was based on LPS-induced PGE2 production model using a homogeneous time-resolved fluorescence(HTRF) PGE2 testing kit combined with liquid handling automation and detection instruments. The critical steps, including the cell density optimization and IC50 values determination of a positive compound, were taken to verify the stability and sensibility of the assay. Low intra-plate, inter-plate and day-to-day variability were observed in this 384-well, high-throughput format assay. Totally 5 121 samples were selected from the company's traditional Chinese medicine(TCM) material base library and used to screen PGE2 inhibitors. In this model, the cell plating density was 2 000 cells for each well; the average IC₅₀ value for positive compounds was (7.3±0.1) μmol; the Z' factor for test plates was more than 0.5 and averaged at 0.7. Among the 5 121 samples, 228 components exhibited a PGE2 production prohibition rate of more than 50%, and 23 components exhibited more than 80%. This model reached the expected standards in data stability and accuracy, indicating the reliability and authenticity of the screening results. The automated screening system was introduced to make the model fast and efficient, with a average daily screening amount exceeding 14 000 data points and provide a new model for discovering new anti-inflammatory and analgesic drug and quickly screening effective constituents of TCM in the early stage.

Objective To examine serum TFAR19 activity in uterine myoma patients.Methods 45 of operation patients with uterine myoma in Beijing Huairou Maternal and Child Health Hospital were recruited.Among them,diameter of maximal myoma from 6 to 10 centineters were 29 (group A),diameter of maximal myoma more than 10 centineters were 16 (group B).In addition,25 unrelated healthy women of childbearing age from physical examination center were enrolled as controls (group C),all of the peripheral venous blood was collected after admission.Enzyme-linked immunosorbent assay (ELISA) was used to examine serum TFAR1 9 activity in uterine myoma patients.Results The activity of TFAR1 9 in group A,group B and group C were 25.32±1.93,14.41±2.28 and 34.79±4.65 respectively.The TFAR19 activity in each group was sta-tistically significant difference (F=7.93,P=0.01).Compared with group C,the TFAR19 activity in group B (q=5.606,P<0.001),group A (q=3.055,P=0.034)was statistically significant difference.The TFAR19 activity in uterine myoma patients subgroups (group A,group B)was statistically significant difference (q=3.086,P=0.033).Age between each groups was not statistically significant difference (F=2.086,P=0.132).Conclusion The decrease of TFAR19 can inhibit apoptosis.Participating the occurrence of uterine myoma,it is connected with diameter of myoma.The larger diameter of my-oma,the lower TFAR1 9 activity of examination will be.

Objective:To establish the quality standard for Naoxin Luotong pills. Methods:Astragali radix, bombyx batryticatus and scorpio were identified under a microscope. Peach kernel, astragali radix, paeoniae radix alba, rheum, paeonia suffruticosa and salvia miltiorrhiza were identified by TLC. The content of total ferulic acid was determined by HPLC with a Sapphire C18 column (250 mm × 4. 6 mm,5 μm). The mobile phase was acetonitrile-0. 1% phosphoric acid(12 ∶88)and the detection wavelength was 316 nm. The column temperature was 35℃ and the flow rate was 1. 0 ml·min-1 . Results:The microscopic characteristics of astragali radix, bombyx batryticatus and scorpio were obvious to be observed easily. The TLC spots of peach kernel, astragali radix, paeoniae radix al-ba, rheum, paeonia suffruticosa and salvia miltiorrhiza were clear and specific. Ferulic acid had a good linear relationship within the range of 5. 1-81. 6μg·ml-1(r=0. 999 9), and the average recovery was 103. 30% with RSD of 1. 96% (n= 9). Conclusion:The standard can be used for the quality control of Naoxin Luotong pills.

Placenta, an important organ, mediates the exchange of nutrients and metabolites between mother and fetus. The transporters, including ATP-binding cassette (ABC) transporters and solute carrier (SLC), expressed in the syncytiotrophoblast play a vital role in substance exchange. Some transporters, such as organic cation transporters (OCTs) and organic anion transporters (OATs), mediate the uptake of endogenous substances and drugs. Some transporters, such as P-glycoprotein (P-gp), breast cancer resistance protein (BCRP) and multidrug resistance-associated proteins (MRPs), can excrete their substrates from the syncytiotrophoblast to the maternal circulation. However, the expression and activity of these transporters are not uniform throughout the gestation period, since they can be affected by physiological and pathological changes during pregnancy or drugs. Thus, an understanding of the role of placental transporters and the variation in their expression and activity in response to physiological and pathological changes is essential for efficient and safe therapy during pregnancy, and it also has important value in the development of drug treatment in pregnancy.
